Ligase Calculation Formula

The following formula is used to calculate the ligase volume required for your NEB ligase reaction.

Ligase Volume = (DNA Concentration * Insert to Vector Ratio + Vector Concentration) / Reaction Volume

Variables:

• DNA Concentration is the concentration of your DNA sample (ng/μL)
• Vector Concentration is the concentration of your vector sample (ng/μL)
• Insert to Vector Ratio is the ratio
